Механизм действия макролидного антибиотика филипина на клеточные и липидные мембраны

Аннотация

В данном обзоре изложены результаты по исследованию механизма действия макролидного пентаенового антибиотика филипина на клеточные и бислойные липидные мембраны (БЛМ). Экспериментально подтверждена «стериновая гипотеза» механизма действия филипина. Изучено гемолитическое и генетико-цитологическое действие филипина, изложены данные действия филипина на вирусные инфекции и на болезни, вызываемые прионовыми белками. Представлены данные исследования механизма действия филипина на БЛМ. При малых концентрациях филипина (1 • 10-8 М) в водном солевом растворе на мембранах с холестерином обнаружены одиночные ионные каналы с проводимостью 15-20 пСм, что примерно в 3-4 раза превышает проводимость амфотерициновых каналов. Обнаружены комбинированные каналы филипина и амфотерицина В, проводимость которых составляет 25-30 пСм, что в 1,5-2 раза выше «чистых» филипиновых каналов и примерно в 5 раз выше «чистых» амфотерициновых каналов. Избирательная проницаемость филипиновых каналов преимущественно катионная.
